Dow Lab Base Editing Sensor Libraries

  • Purpose

    The Mouse Base Editing Sensor (MBES) and Human Base Editing Sensor (HBES) libraries are made up of sgRNAs that target frequently occurring cancer-associated mutations identified in the MSK IMPACT dataset that can be modeled with cytosine base editing. The ALL-PAM sensor (AP) library is made up of 10 human and mouse sgRNAs, where for each sgRNA the tethered target site has 1 of 64 possible 3-bp PAM sequences. Notably these "sensor libraries" also contain the cognate genomic target site within the same expression cassette for quantification of base editing efficiency.
